Right after I've logged in, I can update anything that needs updating, via the Update Manager. After that, I just a get a red cross in the shield and the message 'Could not refresh the list of updates.' Ideas? I've tweaked the sources list but that does nothing.

I'm not familiar with Mint so this is generic advice, however it sounds like there might be some kind of automated update running in the background which is setting a lock for the package manager. If this automated process gets stuck or can't proceed it might just hang and prevent you from doing any updates.
You might want to look at doing updates from the command line to make sure all the packages are properly installed or possibly look at disabling the automatic updates temporarily to see if that fixes it.
